Myalgia, amitriptyline and alternatives

@Allele What dose of PEA do you use? And do you split dose? I've never heard of this stuff before, so I'm intrigued and may research it when I have the energy. A quick check on Google and Amazon suggests it may be too expensive for me, whatever its benefits.

Edit : I found a version of this stuff for animals, advertised under the name :

"Pharmox Dog and Cat PEA"

The name wasn't appealing for some reason. :D

 Arnie Pie you're on a roll today!

I basically take 1 350mg capsule at night, with a daytime dose when I remember (I often forget my daytime doses of everything, even with alerts, am so forgetty.)

I have no idea if this works in combo with the other things I'm taking, but the shift was remarkable soon after I started taking it.
Since it has some action via the cannabinoid receptors, it might be working so well for me bc I also use the raw juice of cannabis (non-psychoactive) which greatly reduces inflammation for me. But if there is or is not a synergistic effect I couldn't say as I am not a chemist or pharmacologist :nerd:

Palmitoylethanolamide (PEA) is an endogenous fatty acid amide, belonging to the class of nuclear factor agonists. PEA has been demonstrated to bind to a receptor in the cell-nucleus (a nuclear receptor) and exerts a great variety of biological functions related to chronic pain and inflammation. The main target is thought to be the peroxisome proliferator-activated receptor alpha (PPAR-α).[2][3] PEA also has affinity to cannabinoid-like G-coupled receptors GPR55 and GPR119.[4] PEA cannot strictly be considered a classic endocannabinoid because it lacks affinity for the cannabinoid receptors CB1 and CB2.[5] However, the presence of PEA (and other structurally related N-acylethanolamines) has been known to enhance anandamide activity by an "entourage effect".[6][7]

Several papers have demonstrated that an imbalance of the endocannabinoid system (ECS) and alterations in the levels of PEA occur in acute and chronic inflammation.[8] For instance during β-amyloid-induced neuroinflammation the deregulation of cannabinoid receptors and its endogenous ligands accompanies the development and progression of disease.[9]

PEA has been shown to have anti-inflammatory,[3] anti-nociceptive,[10] neuroprotective,[11] and anticonvulsant properties.[12]
