It’s because they’ve subset. If you need to get to more specific subsets, you’ll first cluster around two more general markers, draw a box, and then cluster further only on cells in that box. That’s how you get studies saying they sorted something like CD45+CD11b+CD4-CD8-Ly6G-SiglecF-NK1.1-CD64+ cells
