    Alpha 7 (α7)
is a subunit of the neuronal nicotinic receptor that can combine to form homomeric receptors. The α7 nAChR is prominent in the hippocampus, found on GABAergic interneurons of stratum oriens and stratum radiatum, and on pyramidal neurones.

Modulatory effects of α7 nAChRs on the immune system and its relevance for CNS disorders

Notably, the anti-inflammatory response to α7 nAChR stimulation also occurs in the brain. Thus, activation of α7 nAChRs is known to alter the phenotype of both macrophages and microglia from an M1-like to an M2-like phenotype [11, 19, 82, 83]. Consequently, any dysfunction in the α7 nAChR and its signaling processes could tip the balance towards more inflammation.

Although not yet reported, a similar co-agonist effect may occur at α7 receptors expressed by immune cells. If true, this would imply that low doses of a α7 nAChR agonist would be sufficient to enhance the anti-inflammatory response to the endogenous agonists (choline and/or acetylcholine).

The current agonists and positive allosteric modulators of α7 nAChR for CNS indications in clinical trials

The alpha-7 nicotinic acetylcholine receptor (α7 nAChR), consisting of homomeric α7 subunits, is a ligand-gated Ca2+-permeable ion channel implicated in cognition and neuropsychiatric disorders.
