Neurodevelopment Genes Encoding Olduvai Domains Link Myalgic Encephalomyelitis to Neuropsychiatric Disorders, 2025, Lidbury et al

If you have time @wigglethemouse, can you explain a bit about what those warnings mean?

Genetic Predisposition for Myalgic Encephalomyelitis/Chronic Fatigue Syndrome: A Pilot Study 2019 Perez Nathanson Klimas et al

This paper found many problems with this Klimas team paper, confirming @wigglethemouse's concerns. Re-analysis of genetic risks for Chronic Fatigue Syndrome from 23andME data finds few remain, 2021

Neurodevelopment Genes Encoding Olduvai Domains Link Myalgic Encephalomyelitis to Neuropsychiatric Disorders, 2025, Lidbury et al

Thanks for linking that interesting paper @wigglethemouse. I missed it when it came out - here's the forum thread link.

Preprint A Proposed Mechanism for ME/CFS Invoking Macrophage Fc-gamma-RI and Interferon Gamma, 2025, Edwards, Cambridge and Cliff

What is the source of the 80%? The data is of course a mess, with poor characterisation of ME/CFS, especially in those early months. We mentioned before the problem with saying that hardly anyone recovers from ME/CFS is that creates a situation where anyone who reasonably believes that they...

Neurodevelopment Genes Encoding Olduvai Domains Link Myalgic Encephalomyelitis to Neuropsychiatric Disorders, 2025, Lidbury et al

Nevada replication PTPRD That gene is covering an awful lot of ground - even just the first two things mentioned 'cell growth and differentiation' cover an enormous number of possible things that might be going wrong in a wide range of places. And again, there might be a problem caused by...

Neurodevelopment Genes Encoding Olduvai Domains Link Myalgic Encephalomyelitis to Neuropsychiatric Disorders, 2025, Lidbury et al

Yes, I agree. The NBPF genes weren't replicated in the Nevada cohort. I think the attention should go to the genes that they say were found in both cohorts. I think it's possible to make up all sorts of speculative stories. For example, variations in the NBPF genes have been associated with...

    ME/CFS Music

Wonderful, @forestglip, thank you. I haven't managed to listen to many of them yet, but these people are very talented. I enjoyed Sarah's song in post#3 very much, she has a terrific voice. And the German Still Loud song in post#1 is powerful, it had me in tears, also sung by someone with a...

Preprint A Proposed Mechanism for ME/CFS Invoking Macrophage Fc-gamma-RI and Interferon Gamma, 2025, Edwards, Cambridge and Cliff

I agree. I think precision around this issue is important in ME/CFS papers. It is useful if authors make a clear definition of the disease they believe they are talking about. I don't think we have solid evidence that most people who fit the CCC at six months have an illness that never fully...

[...] T cell activation and exhaustion in plasma are associated with persistent symptoms up to 18 months following [COVID], 2025, Ueland et al

Figure 2, where people with more than one persistent symptom are separated out, is a bit more interesting. This is still just of home-isolated participants. but, there's still only a small number of people with elevated levels.

[...] T cell activation and exhaustion in plasma are associated with persistent symptoms up to 18 months following [COVID], 2025, Ueland et al

Sadly, it's not looking like there is much of an effect there. Just possibly, there is an initial elevation of CD25 in a small proportion of people reporting fatigue, but it is gone by 18 months. Perhaps fatigue is just too common a symptom.

[...] T cell activation and exhaustion in plasma are associated with persistent symptoms up to 18 months following [COVID], 2025, Ueland et al

Norwegian study It doesn't look as though there was undue selection of the home-isolated (acute mild) patients for persistent symptoms in this prospective study. Around 50% of the home-isolated patients were men.

United Kingdom: Bath paediatric CFS/Fatigue clinic - Esther Crawley; Phil Hammond

I think that makes it clear that Phil Hammond should not be anywhere near people with ME/CFS seeking medical care.

Agomelatine but not melatonin improves fatigue perception: A longitudinal proof-of-concept study, Pardini et al, 2014

Given this drug works on melatonin receptors, I just want to mention concerns about melatonin that I have mentioned elsewhere. A Professor working in the field of the management of farm animals raised serious concern about the use of melatonin in young people, primarily due to melatonin's use...

Agomelatine but not melatonin improves fatigue perception: A longitudinal proof-of-concept study, Pardini et al, 2014

The safety of agomelatine in standard medical practice in depressed patients: A 26‐week international multicentre cohort study 2020 A large European observational study conducted from 2009 to 2014. It's unclear to me why it took so long to publish. Agomelatine was developed and is marketed by...

Agomelatine but not melatonin improves fatigue perception: A longitudinal proof-of-concept study, Pardini et al, 2014

Agomelatine is a serotonin antagonist, blocking serotonin receptors (so presumably reducing its action), whereas SSRIs block serotonin reuptake, (so presumably increasing the effect of serotonin). Agomelatine appears to be effective in making a range of measures better. The FSS has 9 questions...

Agomelatine but not melatonin improves fatigue perception: A longitudinal proof-of-concept study, Pardini et al, 2014

FSS is Krupp Fatigue Severity Scale. They asked about side effects, and didn't find any difference between the treatment groups in reported side effects. So, perhaps the blinding was effective.

Agomelatine but not melatonin improves fatigue perception: A longitudinal proof-of-concept study, Pardini et al, 2014

Paywall Fukuda criteria I can't see if the study was blinded or not. I'm interested to hear more about the study. It suggests that melatonin in this dose and formulation is not helpful for (ME)/CFS.
