Exploring a genetic basis for the metabolic perturbations in ME/CFS using UK Biobank, 2025, Armstrong et al

I do understand the concerns posted by others. Just to add that given this study, LXR was also identified as being downregulated in Hanson's work as well : https://pmc.ncbi.nlm.nih.gov/articles/PMC10764978/pdf/JEV2-13-12403.pdf Also, even this is unpublished , LXR was one of the first...

Preprint Identification of Novel Reproducible Combinatorial Genetic Risk Factors for [ME] in [DecodeME Cohort] and Commonalities with [LC], 2025, Sardell+

Relevant thread on X. Note the association of CH25H, CYP7B1 (2 of core genes identified) with Epstein-Barr virus https://threadreaderapp.com/thread/1997005812838076483.html

Preprint Identification of Novel Reproducible Combinatorial Genetic Risk Factors for [ME] in [DecodeME Cohort] and Commonalities with [LC], 2025, Sardell+

I am still working in trying to connect the list of genes from this paper with previous research efforts. Some new findings : NLGN1 : Appears on the Snyder study (HEAL2) CYP7B1 : The node of this gene appears on the network analysis (2017) - towards the center bottom : CH25H : Also...

Preprint Identification of Novel Reproducible Combinatorial Genetic Risk Factors for [ME] in [DecodeME Cohort] and Commonalities with [LC], 2025, Sardell+

Comma separated list of the 259 candidate core genes. Nice to see ABCA1 and ABCC6 : Related to ABCA1 and ABCC6 we have the following: ABCC6 https://www.s4me.info/threads/abcc6-and-pathogenic-snps.14251/#post-246766 and ABCA1...

Preprint Identification of Novel Reproducible Combinatorial Genetic Risk Factors for [ME] in [DecodeME Cohort] and Commonalities with [LC], 2025, Sardell+

The gene PDE4B they mention appears also in the genes identified by Snyder et. al : https://www.s4me.info/threads/dissecting-the-genetic-complexity-of-myalgic-encephalomyelitis-chronic-fatigue-syndrome-via-deep-learning-powered-genome-analysis-2025-zhang.43705/

My story: years of ME/CFS is actually a glycogen storage problem

I see that there is mixed evidence about this, @darrellpf is it a homozygous or heterozygous mutation ? even with a heterozygous one -which was thought to be benign- there is evidence that it may not be.

My story: years of ME/CFS is actually a glycogen storage problem

Congratulations on your efforts. Make sure you crosscheck with other AI engines the answers. My suggestion is the use the MOA (Mixture of Agents) function by Genspark AI. Every recovery story gives valuable data points. I have shared with a researcher the attached file, which lists a number of...

Can Large Language Models (LLMs) like ChatGPT be used to produce useful information?

I have no doubt that the reasoning process of LLMs can help put pieces of the puzzle of ME/CFS together. Unfortunately people having seen hallucinations believe that *every* output from LLMs is total crap. I recall reading someone saying "as soon as I read that something is from an LLM, I stop...

Cellular Nitrogen and Energy Metabolism in ME/CFS, 2025, Armstrong et al

I second that @MelbME . I recently came across an ME patient with Fibromyalgia. She was found to have a problem with gene ALDH18A1, type SPG9A. She is now on a specific diet (not too much protein) and supplements with L-Citrulline. She is able to do more and now does not experience any spasms...

Temporal dynamics of the plasma proteomic landscape reveals maladaptation in ME/CFS following exertion, 2025, Germain et al.

Once again, I see how potentially important information may go unnoticed. I am putting here just an example cc @DMissa Among the genes observed we have CBLN4. So I plugged this gene to the Information retrieval system I have been using : Here are the rankings observe that N-Linked glycosylation...

    Machine Learning-assisted Research on ME/CFS

I wanted to have this here given my previous work, a run which was performed 10 years ago which identified potential research targets for MECFS cc @forestglip @ME/CFS Science Blog : Source ...

Preprint Virus Genome Sequences in the Blood of Myalgic Encephalomyelitis/Chronic Fatigue Syndrome Patients, 2025, Davis et al

Of course it may well be what @ME/CFS Science Blog suggests : Fewer contacts of patients means that the probability of infection is very low. However, I would like to mention a subset of ME/CFS patients who say that ever since they gotten ME/CFS they do not get viral infections at all. I wonder...

Preprint Initial findings from the DecodeME genome-wide association study of myalgic encephalomyelitis/chronic fatigue syndrome, 2025, DecodeMe Collaboration

Thanks @forestglip , yes I wonder if a key mechanism here is excitotoxicity via glutamate (and perhaps quinolinic acid). Another interesting finding can potentially be the gene ACO2 (exists also in the list of genes) which could be linked to the itaconate shunt hypothesis. If we have indeed...

Preprint Initial findings from the DecodeME genome-wide association study of myalgic encephalomyelitis/chronic fatigue syndrome, 2025, DecodeMe Collaboration

@forestglip I am not sure if you have seen this analysis by Paolo Maccallini : https://github.com/paolomaccallini-hub/MetaME?tab=readme-ov-file I am particularly interested in genes EP300 and UGP2. Can these two be significant targets?

Systemic increase of AMPA receptors associated with cognitive impairment of long COVID, 2025, Fujimoto et al.

Thanks to user Violeta for pointing this out. In one of my algorithmic runs, glutamate has been identified as a key factor for MECFS symptoms. Using the HotSpot algorithm and having the target feature 'glutamate' AMPA has been identified as shown below [1] : It is interesting to see that...

Proximity to Golf Courses and Risk of Parkinson Disease, 2025, Krzyzanowski et al

I went through the links which mentioned that paraquat could be a herbicide that is responsible for an increased probability for getting Parkinsons. So what is the mechanism of action of paraquat? Oxidative stress, lipid peroxidation, cell membrane disruption. That rings many (3) bells .
