The Occurrence of Hyperactivated Platelets and Fibrinaloid Microclots in ME/CFS, 2022, Nunes, Pretorius et al

I'd imagine it's pretty hard to get a sense of physical density looking at fluorescent markers, but for what it's worth these images of larger structures from acute Covid patients in their previous paper look fairly loose. What they saw in ME samples, in comparison, was smaller and less...

The Occurrence of Hyperactivated Platelets and Fibrinaloid Microclots in ME/CFS, 2022, Nunes, Pretorius et al

Wouldn't this depend on their density and form? Platelets are presumably much denser than these fibrin polymers, which as I understand it are more like a network in the plasma than discrete objects like platelets. So a centrifuge that would spin out platelets wouldn't necessarily spin out 'micro...

Clinical overlap between fibromyalgia and myalgic encephalomyelitis. A systematic review and meta-analysis, 2022, Ramírez-Morales et al

My impression from a very small sample is that doctors in Mexico don't generally know anything at all about ME. Maybe that will change with somewhat growing awareness of Long Covid. It looks like these authors all have experience with FM and work in rheumatology, and don't seem to have much...

The Occurrence of Hyperactivated Platelets and Fibrinaloid Microclots in ME/CFS, 2022, Nunes, Pretorius et al

They definitely weren't blinded for the platelet spreading or clumping they visually assigned scores of 1-4 to though, as they don't even report values for controls, so it appears they only scored ME platelets.

Long Covid in the media and social media 2022

In the Pipeline blog: Long Covid, Long Other Things Derek Lowe's blog (which is generally an insider perspective on the pharma industry) is usually quite good, but this one, which takes a somewhat skeptical view of LC, ME, etc, is pretty frustrating. He also took a pretty positive look at the...

Mental health and alcohol use among patients attending a post-COVID-19 follow-up clinic: a cohort study, 2022, Broughan et al.

They say in the paper that the 45.5% they observed at 3 months is similar to the general population in Ireland (which is actually 52%). The one year data is just wrong and I'm in touch with them to get it corrected.

The Occurrence of Hyperactivated Platelets and Fibrinaloid Microclots in ME/CFS, 2022, Nunes, Pretorius et al

I also wonder about past Covid being an issue with these results. They say that patients were excluded if they had past Covid infection, but they don't say how and they don't report any serology results (though even that would not be definitive). I imagine in 2022 in South Africa where infection...

The Occurrence of Hyperactivated Platelets and Fibrinaloid Microclots in ME/CFS, 2022, Nunes, Pretorius et al

Looking at their past papers, looks like they are claiming fibrin amyloid formation is present in migraine, T2 diabetes, rheumatoid arthritis, Alzheimer's, sepsis, septic shock, SIRS, and multiple organ dysfunction syndrome, acute Covid, Long Covid and now ME. That's just going off the titles of...

The Occurrence of Hyperactivated Platelets and Fibrinaloid Microclots in ME/CFS, 2022, Nunes, Pretorius et al

I'm not sure what's going on, but it doesn't look like these are directly comparable. The controls in that earlier paper showed about five times higher average amyloid area %. So maybe the method was somehow different or maybe the control group was different.

The Occurrence of Hyperactivated Platelets and Fibrinaloid Microclots in ME/CFS, 2022, Nunes, Pretorius et al

If this turns out to be a solid finding, I wonder how much we'll see it in other diseases as well. The same authors have previously done work that looks at least superficially similar in rheumatoid arthritis. It might be a quite general finding in the end. Regarding the first experiment: "A...

The Occurrence of Hyperactivated Platelets and Fibrinaloid Microclots in ME/CFS, 2022, Nunes, Pretorius et al

Not a direct replication, but the Roy et al paper on ATG13 did find similar results, also showing quite a strong effect, in a small sample (7 patients and 7 controls) using Thioflavin T as well. This is pretty similar to the first part of what Pretorius did in this paper (though not the addition...

The Occurrence of Hyperactivated Platelets and Fibrinaloid Microclots in ME/CFS, 2022, Nunes, Pretorius et al

Abstract We have previously demonstrated that platelet poor plasma (PPP) obtained from patients with LongCovid/Post-Acute Sequelae of COVID-19 (PASC) is characterized by a hypercoagulable state reflected in hyperactivated platelets and the presence of considerable numbers of fibrin(ogen) amyloid...

Long Covid in the media and social media 2022

The data is definitely and obviously bad. It's not even an asking the wrong questions, it's just bad data collection and bad calculations. I'm in touch with the author and the editor to try to get this fixed.

Yoga is effective in treating symptoms of Gulf War illness: A randomized clinical trial Hayley,Younger et al 2021

Someone needs to invent sham yoga, so these trials have something to use as a placebo treatment. Just a random sequence of randomly-generated body positions at the same tempo as a yoga session, but with the same vaguely spiritual veneer.

Mental health and alcohol use among patients attending a post-COVID-19 follow-up clinic: a cohort study, 2022, Broughan et al.

It is very easy to score mild or moderate on the PHQ-9 if you have Long Covid without being depressed at all. Being tired and having some brain fog puts you in the mild category, add in some difficulty sleeping and a little moving slowly and you're moderate.

Mental health and alcohol use among patients attending a post-COVID-19 follow-up clinic: a cohort study, 2022, Broughan et al.

I downloaded their data and had a look at some of the details and the drinking data definitely doesn't add up. I don't think we can trust any of their alcohol conclusions and I'd want to look very carefully at the other data as well before believing it (but I've gone past my limit for this kind...

Mental health and alcohol use among patients attending a post-COVID-19 follow-up clinic: a cohort study, 2022, Broughan et al.

Also the depression scores went down very slightly and the anxiety scores went down significantly between 3 months and a year post-Covid, while alcohol use went up. So probably not the case that worsening mental health is driving more drinking.

Mental health and alcohol use among patients attending a post-COVID-19 follow-up clinic: a cohort study, 2022, Broughan et al.

As far as I can tell, a person drinking a glass of wine with dinner twice per week would qualify as having problematic alcohol use in this study (as would someone who was drinking to excess daily). The data is available, so someone could actually do a more detailed analysis. Is alcohol...

News From Jarred Younger / Neuroinflammation, Pain, and Fatigue Laboratory at UAB, From Aug 2020

Solve M.E. webinar, June 23: How We Can See ME/CFS Inflammation in the Brain Dr. Jarred Younger’s 2016 Ramsay Award was used to show that brain temperature is elevated in ME/CFS. This increased temperature is a sign of brain inflammation. The findings are now being replicated in a large group...
